课题组于2008年11月成立,系统研究表观转录组调控与RNA甲基化功能机理,在拓展RNA甲基化表观转录组调控的前沿新领域合作研究中,发现人源 RNA m6A修饰甲基转移酶复合物METTL3/METTL14/WTAP(Cell Res 2014 年,为 Natl Sci Rev 研究特色论文),揭示非编码miRNA介导的m6A甲基化位点形成的选择性机制(Cell Stem Cell 2015,入选当期封面特写报道和 Cell 出版社的研究特色论文),参与鉴定m6A去甲基化酶ALKBH5(Mol Cell 2013,作为 RNA 生物学研究前沿热点入选 Science Editor’s Choice)和FTO(Nat Chem Biol 2011),发现m6A修饰作为mRNA新顺式元件调控mRNA剪接和翻译功能(Cell Res 2014,为 Cell Res 研究特色论文;Mol Cell 2016,为Trends In Genetics Hotspot论文;Cell Res 2017),揭示m6A调控造血干细胞定向分化和精子发生减数分裂机制 (Nature 2017; Cell Res 2017, 入选当期封面特写报道);还发现了m5C修饰通过其结合蛋白ALYREF调控mRNA出核机制(Cell Res 2017, 入选当期封面特写报道)。这些结果表明RNA甲基化修饰具有可逆性并调控RNA加工代谢作用,拓展了RNA甲基化表观转录组前沿性领域,并应邀撰写多篇综述(Genom Prote Bioinfor 2013; Curr Diab Rep 2014; Sci Bull 2015; RNA Biol 2016)。
课题组研发RNA化学修饰高通量测序和分析技术,系统研究RNA甲基化及其修饰酶,阐明其功能及遗传性状(表型)和疾病关联的调控机制。

RNA转录调控与发育及环境应答机制
合作团队绘制了首个PM2.5诱导的肺纤维化中RNA m5C图谱,发现m5C增益的上调基因可能对PM2.5诱导的小鼠肺纤维化发展至关重要,对开发PM2.5暴露相关性肺病的潜在生物标志物具有潜在价值(Han et al. Genomics Proteomics & Bioinformatics 2020);鉴定了水稻mRNA m5C甲基转移酶OsNSUN2,构建了甲基化图谱,发现m5C修饰调控mRNA翻译效率,发现36 条件下osnsnu2突变体的叶绿体功能紊乱,导致光依赖的活性氧积累,揭示OsNSUN2介导水稻mRNA的m5C修饰,调控水稻高温适应性(Tang et al. Developmental Cell 2020);从代表性RNA修饰(m6A、m5C等)特征规律切入,系统综述RNA甲基化参与多种癌症的致病机理(Han et al. Seminars in Cancer Biology 2020);绘制了包含RNA四个碱基信息的斑马鱼早期胚胎发育RNA二级结构全景图谱,并发现RNA结构影响结合蛋白Elavl1a对RNA结合能力,从而调控母源mRNA稳定及降解,调节斑马鱼母源-合子转换及早期胚胎发育进程(Shi et al. Genome Biology 2020)。
