报告题目: 肿瘤驱动基因网络的发现、鉴定及其机制研究

报告摘要:
肿瘤的发生与演进起源于遗传和表观遗传的异常变化。这些变异导致癌基因的激活、抑癌基因的失活、以及其他分子和信号通路的变化,使正常细胞发生转化、癌细胞进一步恶化。发现、鉴定肿瘤驱动基因并研究它们的相关信号通路、分子网络及调节机制对于改善癌症的检测、预防与治疗发挥着决定作用。利用遗传、生化、分子细胞生物以及功能检测的各种手段和细胞、小鼠以及人体组织等各种研究材料,我们多年致力于肿瘤基因的发现和功能、机制方面的研究。先后发现了KAI1(抗癌1号)肿瘤转移抑制基因、KLF5和ZFHX3抑癌基因以及ERRF、U50、FOXA1、WWP1等多个肿瘤基因。我们还对KLF5和ZFHX3进行了较深入的功能和机制方面的研究,发现KLF5通过作为TGFβ信号通路的一个关键因子实现其抑癌作用、KLF5乙酰化状态决定KLF5和TGFβ发挥抑癌功能还是促癌功能、ZFHX3通过诱导细胞分化来实现其抑癌功能、ZFHX3与激素信号相互作用并调节上皮细胞分化。将基因组和蛋白质组学手段应用到肿瘤材料将导致对肿瘤的各种生物学行为的深入全面的了解,从根本上改变对肿瘤的检测、预防、药物开发以及治疗方案的制定。
报告人简介:
Dr. Jintang Dong has expertise in cancer genetics and biology that is helpful for the success of this proposal. Both of his training and independent research have been on these areas. By publishing more than 100 research articles, he has the expertise in multiple areas of cancer biology research, including the discovery and characterization of cancer genes; functional evaluation of cancer genes in various models such as in vitro cultured cells, xenograft tumor models and genetically modified mice; understanding of how cancer genes function in both normal tissues and tumor cells; how genes are transcriptionally regulated; and how cancer genes regulate the balance of cell proliferation and differentiation in epithelial homeostasis. He has also been involved in head and cancer research at Emory, as he has been the Director for the Developmental Research Program in the Emory SPORE in Head and Neck Cancer. He has mentored multiple junior faculty members during his career so far.
